Abstract (EN)
In developed countries, life in the process of development of colorectal cancer rate is 5%. it is the second leading cause of death from cancer. The presence of family history of colorectal cancer is 25-35% and it is a well-known risk factor. Apoptosis is called as a programmed cell death. The unwanted cells in an organism are eliminated to maintain equilibrium. In this study, fas ve trail genes are associated with apoptosis in their relation with colon cancer were investigated. In this study, a polymorphism of the Trail gene at position 1595 exon 5 and Fas gene at position 1377 promotor region were examined in this study. Firstly paraffin tissue samples were taken from colon cancer individuals and blood samples were taken from healthy individuals. Then DNA was isolated using by kit. These polymorphisms were investigated using PCR-RFLP methods. The findings were evaluated by the appropriate statistical methods. We have examined Fas -1377 and Trail +1595 polymorphisms as a marker of tumour risk and progression in colon cancer. 56 patients with colon cancer and 46 healty controls were enrolled in this study. According to our results, There were found a significant association in the distribution of Fas -1377 genotypes and Trail +1595 genotypes and their alleles between patients with colon cancer and controls. Fas -1377 AG genotype (p=0.001) and Trail +1595 CT (p=0.015) genotype in patient were seen more than those in controls and than other genoype. There were no association between sex, tumor grade and type, family history, cigarette and alcoohol at Trail or Fas genotypes.These findings if confirmed would have clinical value in helping to assess the genetic risk and tumour progression for brain tumours thus opening new perspectives for the study of molecular factors underlying the mechanisms of colon cancer.
