Abstract (EN)
Kidney failure is a worldwide public health problem with its increasing incidence and prevalence, high costs and poor outcomes. Disease and management are classified according to stages of disease severity and clinical diagnosis, as assessed by glomerular filtration rate (GFR) and albuminuria. Chronic kidney disease is a disease that can be detected by routine laboratory tests, and some treatments can prevent its progression and slow the progression of the disease. Therefore, it is important to discover new biomarkers for the diagnosis and follow-up of the treatment of the disease. The aim of this study; To examine the plasma free amino acid profiles both before and after dialysis in patients with chronic kidney disease and to investigate the potential for use in diagnosis and treatment by comparing them with the healthy control group. In our study, plasma samples were obtained from 46 healthy patients diagnosed with CRF and 46 patients diagnosed with CRF who applied to Şanlıurfa Harran University Medical Faculty Dialysis Department. Plasma free amino acid profiles were analyzed by LC-MS/MS method. Statistical analysis of the obtained data was done with SPSS. As a result of statistical analysis, average alanine, arginine, aspartic acid, citrulline, histidine, methionine, tyrosine and hydroxyproline glycine, leucine, isoleucine, lysine, ornithine, phenylalanine, proline, serine glutamic acid, glutamine valine, taurine alloisoleopic acid in patients with chronic renal failure, , anserine, gammaaminobutyric acid, 1- methylhistidine, 3-methylhistidine 5-oh-trp amino acid levels were found to be higher than the control group. Phosphoethanolamine, cystine, alphaaminobutyric acid, betaaminobutyric acid and tryptophan amino acids were found to be lower in CRF patients than in the control group. After dialysis compared to pre-dialysis, citrulline, histidine, alanine, arginine, aspartic acid, glutamic acid, glycine, cystine, isoleucine, proline, phosphoethanolamine, taurine alloisoleucin, alphaaminoadipic acid, ancerine, alphaaminobutyric acid, betahylamine, 1-beta- ohalanin, 1 While there was an increase in -trp level, a decrease was observed in glutamine, leucine, lysine, ornithine, phenylalanine, serine, valine, asparagine, methionine, tryptophan, tyrosine and hydroxyproline gammaaminobutyric acid 3-methylhistidine levels. Citrulline, glycine, lysine, anserine, alphaaminobutyric acid, gammaaminobutyric acid, phosphoethanolamine and taurine levels were found to be significant in the Paired samples T test, which was used to test the significance of the difference between the arithmetic means of the groups (p<0.05). As a result of the study, more studies were needed to understand the role of amino acids in CRF.
