Abstract (EN)

In our study, the possible neuroprotective effects of Quercetin were investigated on mouse neuroblastoma cell line (Nb2a). Quercetin was dissolved in solvent (DMSO, 0.2%) and diluted with medium before using the stock solution. The toxic effects of Quercetin were examined by Neurotoxicity Screening Test (NTT) and cell viability and proliferation test. In the NTT test, neurotoxicity was induced by adding 25 µM concentrations of chlorpyrophos (CPS) to the cell culture. Quercetin was added at concentrations of 1, 3, 10, 30, 50 and 100 μM and its effect on neurite inhibition was examined. The CellTiter-Glo® luminescence cell viability measurement method (Promega, Madison, WI) was used to evaluate the cytotoxicity of quercetin. In order to evaluate the effect of quercetin on cytotoxicity and cell proliferation, mouse neuroblastoma cells (Nb2a) were incubated with 1, 3, 10, 30, 50 and 100 μM concentrations of quercetin and compared with the control group without quercetin and solvent (DMSO, 0.2%). Statistical analyses of the results obtained were performed and p<0.05 values were considered statistically significant
