Abstract (EN)
Purpose: Ghrelin is a peptide hormone which produced by gastrointestinal system has effects on eating behavior and weight regulation; growth hormone (GH) secretion, nutrition, gastric acid secretion, gastric motility and cell proliferation. Burn injury is a major problem affecting mortality and morbidity.In this study, we aimed to investigate the effects of systemic and localy administered ghrelin on wound healing. Materials and Methods: Fourty Sprague Dawley rats were divided into 4 groups. In Burn group, burned rats did not receive any treatment. In Ghrelin-S group, Ghrelin was administered intraperitoneally 20 mg/kg/day to burned rats. In Chitosan group, chitosan gel was applied once a day to the burn wound on rats. In ghrelin-L group, ghrelin with chitosan (gel form of ghrelin) was applied once a day to the burn wound on rats. Biopsy was takenat the 7th and 14th days. The rats were sacrificed on the 14th day. Hydroxyproline level in tissue samples was measured biochemically. In the histopathological examination, mononuclear cell infiltration, epidermal and dermal necrosis and haemorrhage - congestion was evaluated. Results: In the Ghrelin-L group, HP levels was found to be significantly higher than the control group and also mononuclear cell infiltration necrosis and hemorrhage-edema scores were found to be lower than the control group.(p<0,05) Chitosan group also found a positive effect on wound healing but not as effective as the Ghrelin-L group. Systemic ghrelin application has less effect on burn wound healing compared with local aplication. Conclusion: Ghrelin has a positive effect on burn wound healing. Local administration of ghrelin have been found to be more effective than systemic administration in experimental burn model.
