Abstract (EN)
Tıtanıum nanoparticles and alloys are extensively utilised in multiple fields including aerospace, chemical industry, national defense and military and this fact brings metallic Ti to a significant strategic entity. There has been little work conducted on biological and chemical parameters of these nanoparticles so far. That is why, it has been aimed to determine some biochemical effects of Titanium NP in the yeast growth environment and its effects on the defense system in this study. YEDP medium has been made up for the advancement and proliferation of the S. cerevisiae yeast cell to be employed in the experiment. There has been formed experimental group which contains 100 mg/L and 250 mg/L concentrations of Titanium in the growth medium. After the sterilization process, each group of culture samples, which were brought to a temperature suitable for laboratory conditions, were inoculated with the culture of S. cerevisiae cells. Following inoculation process, it was left to incubation. Right after this procedure, the cells were gathered by measuring and centrifuging according to cell densities. The collected cells were homogenized with a mixture of Tris HCl-base and EDTA, and after centrifugation, the supernatant part was homogenized with an HPLC device for GSH (Elman reagent), total protein (Lawry) and TBARS measurements, and the remaining pellets were homogenized with an n-hexane/isopropanol mixture and fatty acids and sterols (ergosterol, campestrol, stigmasteol, betasitosterol) were analyzed. Eventually, it has been observed that Titanium NP Yeast has distinct effects on the biochemical and defense system. An excessive dose of Titanium NP has been exposed to stress in the organism. Last but not least it is thought that the results revealed on the antioxidant defense system will be a source for resembling studies on other biological models, which supports future studies and contributes to the literature.
