Pressurized hot water extraction of bioactive compounds from menengic seed (pistachia terebinthus)

Abstract (EN)

In this study, the pressurized hot water extraction system was designed and bioactive compounds in the contents of the extracts of the menengıc were investigated. The experimental conditions have been optimized for extracting bioactive properties by using Response Surface Methodology (RSM) and Central Composite Design (CCD) technique. The optimum conditions for the total amount of phenolics were determined as 17.37 mg gallic acid / ml extract concentration, 100 oC temperature, 3 ml / min flow rate and 10% solid liquid ratio and the maximum gallic acid concentration was obtained as 15.297 mg gallic acid/ml under this conditions. The optimum conditions for the total amount of flavonoids were determined as 73,07 µg quercetin/ml extract concentration, 193 oC temperature, 3 ml / min flow rate and 6,21% solid liquid ratio and the maximum quercetin concentration was obtained as 78,803 µg quercetin/ml under this conditions. The optimum conditions for the DPPH radical scavenging effect determination were determined as 100,29 µmol trolox/ml extract concentration, 150 oC temperature, 2,52 ml / min flow rate and 8,53% solid liquid ratio and the maximum trolox concentration was obtained as 97,559 µmol trolox/ml under this conditions. Verification experiments were also carried out at the points obtained as a result of optimization of model expressions. The concordance between the results of the model and the experimental results was between 87% and 97%. It was determined that a selective extraction process can be performed at different temperatures with the same filler in the pressurized hot water extraction. In the study, it was also seen that the flow rate should be increased in the new design to be formed in a wider temperature range.
