Abstract (EN)

Mesenchymal to epithelial transition (MET) is a key event that is observed in many physiological processes involving development, cancer progression and tissue regeneration. MET regulation is achieved by many transcriptional events that are dependent on genetic and epigenetic factors. According to recent studies, MET regulation is not exactly the opposite of epithelial to mesenchymal transition (EMT) by virtue of removal of the factors involved in the regulation of EMT. Despite the fact that MET is not very well understood, there are some genes that are thought to take important roles in MET regulation. Elf3 is thought to be essential in the regulation of MET by interacting with the promoter of Grhl3. Moreover, Grhl3 has been reported to act on the cis regulatory element that is found on Cdh1 and it is thought to be associated with E-cadherin localization. In this study, absence of Elf3 is hypothesized to cause alterations in transcriptome dynamics, in terms of coding, noncoding and repetitive transcriptome that presumably involve important factors such as Grhl3. Therefore, differential expression analysis resulted in alterations in the expressions of various of protein coding genes, long noncoding RNAs and repeat elements. Consequently, enrichment analyses involving Gene Ontology and KEGG pathways for differentially expressed genes showed that the absence of Elf3 during MET may lead to disruptions in cell cycle, more particularly DNA replication, mitotic nuclear division and chromosomal segregation.
