Abstract (EN)

Unlike the staining techniques in biochemistry, like Hematoxylin and Eosin (H&E) staining, this thesis demonstrates that biological compartments of different mouse tissues, namely the placenta, and uterus, can be visualized using micro-Raman spectroscopy without labeling. By comparing the H&E stained tissues with the Raman cluster maps, it is successfully verified that the functional zones of mature mouse placental tissues can be realized exclusively using Raman spectroscopy and clustering analysis. Through the spectral analysis, their biochemical differences are also discussed. Additionally, it is shown that the choice of the dimensional reduction and the clustering algorithm has an impact on the performance of the final clustering in the analysis. It is illustrated that t-SNE and DBSCAN algorithms provided superior separability in the scatter plots of the two components compared to the other combinations among 25. Once the combination of algorithms was determined, it was applied to analyze both the tissues taken from implantation and inter-implantation sites. The results demonstrated that uterine tissues taken from implantation sites on day five exhibited a higher density of certain biochemicals near the region where the embryo attaches, which is in line with the histological findings.
