Abstract (EN)
In this thesis, sampling was performed form 14 points between May and September 2014-2016 in order to determine the Culex pipiens species complex which is spreading in the Middle Black Sea Region. A part of the Ace-2 gene region had been amplified in order to determine the species status related to the morphological identification problems in the Culex pipiens species complex. The CQ11 region was reproduced and molecular diagnostics were attempted for determining Culex pipiens varieties which are heavily distributed in the field. The morphologically identified complex members had been used in molecular studies. The amplicons obtained as a result of the PCR reaction were subjected to the sequencing process. Sequence chromotograms were examined and then aligned with Mega7 program and phylogenetic trees were drawn by adding them to the samples taken from the gene bank. According to the obtained data, it was determined just only Culex pipiens species in the area and two varieties of Culex pipiens (Culex pipiens and Culex pipiens f molestus) distribution. Culex pipiens quinquefascitus, which is known to exist in our country and has recently been molecularly confirmed, and Culex torrentium, the twin species of this complex, have not been found in the study area. The drawn UPGMA tree only showed the presence of Culex pipiens f molestus variety at the Samsun Horhor stream. It has been identified each two varieties were exist in Amasya, Bafra, Giresun, Çarşamba, Ordu Ünye and only the presence of the Cx. pipiens pipiens variety in the other regions. As a result of comparison with the gene bank samples, It was found that our samples highly overlapped with KM922631.1 Culex pipiens samples. Culex pipiens quinquefasciatus Gene Bank sample was found to be less distant from Culex pipiens pallens Gen Bank sample, and Culex torrentium was found to be the most distant species.
