Synthesis of pyrimidine derivative compounds from chalcone by microwave method and biological activities

Abstract (EN)

In this study, substituted 4,6-diphenylpyrimidene compounds (185-211, 213-214 and 216-232) were synthesisesd from methoxyl and hydroxyl substituted chalcones (164-184) with urea and thiourea using solid-phase or solvent condition by microwave techniques. Compounds 212 and 215 were synthesisesd from methoxy substituted acetophenone, benzaldehydes and urea in one-pot reaction by microwave reaction. Synthesized pyrimidine compounds 185-211, 212-218, and 219-232 were named as 4,6-diphenylpyrimidine-2-ol/thiol, 4,6- diphenylpyrimidine-2-(1H)-one/thione, and 4,6-diphenyl-3,4-dihydropyrimidine-2(1H)-thione, respectively. 48 Type of pyrimidine compounds were synthesised and compounds 201, 215, 217, 226 and 229 were known in the literature, but rest of compounds 185-200, 202-214, 216, 218-225, 227-228 and 230-232 are not known. Identification of synthetic compounds were made by spectroscopic methods using NMR (1H-NMR, 13C/APT, COSY), FT-IR, UV-VIS, mass (LC-MS/MS, LC/MS-TOF) and elementel analysis techniques and by the help of ACD-NMR spectra. In the study of biological activity evaluation of synthesized compounds 185-232; the most antimicrobial active compounds against gram positive bacteria were 197 and 207- 209 with in the range of 2-9 µg/mL, the better alpha-glucosidase enzyme activity were observed for the compounds 186, 188, 195, 199, 208 and 211 with the value of 3.15 ± 0.60; 6.10 ± 1.31; 2.36 ± 1.18; 3.09 ± 0.42; 3.38 ± 1.36; 4.38 ± 0.36 µM, respectively, and the best lipase enzyme inhibition activity were obtained from compounds 187, 189 and 211 with the value of 0.26 ± 0.07; 0.40 ± 0.06; 0.29 ± 0.02 µM, respectively..
