Abstract (EN)

In this study, laccase enzyme (L) was immobilized using entrapment method in poly(acyrilamide)-sodyum alginate, P(AAm)-NaAlj, poly(acrylamide-N-isopropylacrylamide)-sodyum alginate, P(AAm- NPA)-NaAlj and poly(acyrlamide-N-isopropylacrylamide), P(AAm-NPA) hydrogels. Optimum pH was determined as 5,0, 6,0 and 5,5 for free laccase and enzymes which are immobilized in P(AAm)-NaAlj, P(AAm- NPA)-NaAlj, P(AAm-NPA) hydrogels, respectively. Optimum temprature was determined as 40 oC for free laccase and immobilized laccases. After 56 days of storage at 4 oC, free laccase, P(AAm)-NaAlj, P(AAm-NPA)-NaAlj and P(AAm-NPA) immobilized enzymes retained 42 %, 79 %, 86 %, and 91 % of their original activities, respectively. P(AAm)-NaAlj, P(AAm-NPA)-NaAlj and P(AAm-NPA) immobilized enzymes were retained 71 %, 84 % and 77 % of their original activities when they were used repeatedly 10 times. v Km and Vmax were calculated as 6,7x10-3 mM, 6,2x10-2 mM, 1,9x10-2 mM, 1,0x10-1 mM and 1,8x10-3 mM.dak-1, 1,7x10-2 mM.dak-1, 6,3x10-3 mM.dak-1 2,9x10-2 mM.dak-1 for free and P(AAm)-NaAlj, P(AAm-NPA)-NaAlj, P(AAm-NPA) immobilized laccase.
