Abstract (EN)
Polymer-bound Schiff-base ligands were prepared from (aminomethyl)polystyrene (APS) and thiophene aldehyde (Tio), 5-chloro thiophene aldehyde (5-ClTio) and 5-methyl thiophene aldehyde (5-CH3Tio). Firstly, all of the modified polymers were characterized on the basis of, FT-IR and GPC spectral analyses studies. The changes in the surface structure of modified polymers were examined with SEM-EDX analysis. After that, Glucos oxsidase were immobilized onto (APS-Tio), (APS?5-ClTio) and (APS-5-CH3Tio) modified polymer with using the hydrogen binding. The influence of temperature, pH and reusability, storage capacity on immobilized Glucos oxsidase were investigated. The immobilized glucose oxidase on (APS- Tio)-GOx showed two optimum pH values that were pH = 4.0 and pH = 7.0. (APS-5ClTio)-GOx and (APS-5-CH3Tio)-GOx were showed optimum pH values as 8.0 and 6.0, respectively. (APS-Tio)-GOx was showed optimum temperature as 30 oC and 50 oC at pH 4, and 30 oC and 70 oC at pH 7. (APS-5-ClTio)-GOx and (APS-5-CH3Tio)-GOx were showed optimum temperature as 70 oC (respectively pH 8 and pH 6). Vmax kinetic parameter was calculated as 0.0409 mM-0.6678 mM-0.4052 mM- 1.073 mM/ 0.0259 mM.min?1 (pH 4.0; t=50 °C - pH 7.0; 30 °C- pH 7.0; 70 °C for APS-Tio). Vmax kinetic parameter was calculated as 0.5485 mM for (APS-5- CH3Tio). Vmax kinetic parameter was calculated as 0.5485 mM for (APS-5- ClTio).
