Investigation of effects of profenofos and Mcpa on testis on ultrastructural level

Abstract (EN)
Endocrine disruptors disrupt normal functions of hormones in the body by imitating them or by blocking them. They change normal levels of hormones and stimulate the production of hormones or give pause to their production or disrupt the ways of translocation of hormones in the organism. A significant group of endocrine disruptors is pesticides. The most widely used is group of organophosphate among pesticides. It is known that of these compounds, profenofos and MCPA (4-chloro-2-methylphenoxy acetic acid ) have negative effects on male genital system. However, studies about the effect on the ultrastructural level are limited. For this reason, in this study, investigating the effect of both MCPA and profenofos on testis seminifer tubuls on the ultrastructural level comparatively is intended. In the study, there are three groups and in each group there are ten male rats which are fourteen weeks old. Group 1 is the group of control. Group 2 is the group of profenofos and group 3 is the group of MCPA. Only distilled water is administered to the control group by gavage. Two times per week, on determined day and time, 17 mg/kg profenofos is administered to group of profenofos by gavage for one month. Every day, on determined time 190 mg/kg MCPA is administered to group of MCPA by gavage for one month. All of the rats in the study were sacrificed and tissues of testis were excluded. Electron microscopic and biochemical investigation were made on excluded tissues of testis. According to electron microscopic investigations, the process of spermatogenesis is healthy in the group of control. Structural degenerations were observed on spermatogenic cells and Sertoli cells in the group of profenofos. In the group of MCPA, gaps between cells which make up spermatogenic series and cellular degeneration, structural damage are more clear than profenofos. In the group of MCPA, apoptosis is determined in some Leydig cells. According to biochemical investigations, free and total testosteron levels measured and in the groups of profenofos and MCPA, there are significant differences statistically on the levels of free and total testosteron before and after administration. As a result of structural degeneration observed in the view of testis and biochemical data, we think that profenofos and MCPA impede the process of spermatogenesis to continue healthily.
