Abstract (EN)

In this proposed thesis study, protein-immobilized cryogels will be used for the selective recognition of L-Tryptophan (L-Trp), which is physiologically important and an essential amino acid. The material will be prepared to determine the selectivity and concentration of L-Trp using human serum albumin (HSA) immobilized poly(HEMA-GMA) cryogels. In this context, HSA-immobilized cryogels will be synthesized; after physical and chemical characterization, optimization studies will be carried out to determine L-Trp selectivity. For this purpose, L-Trp adsorption from HSA-immobilized poly(HEMA-GMA) cryogels will be evaluated in terms of pH, initial concentration, ionic strength, and temperature. However, to examine the specific binding relationship between HSA and L-Trp, cryogels such as D-Trp, D, L-Trp, L-Tyrosine, and L-Phenylalanine will be used in selectivity experiments. At the end of this study it is aimed to introduce affinity columns to the literature for use in the recognition and determination of L-Trp concentration.
