Determination of some toxicokinetic parameters of atractyloside in rats

Abstract (EN)
Atractyloside (Atr), which is a traditional plant component consumed by both human beings and animals worldwide, is widely grown in our country, near the stream, in empty fields and near dumps. Especially in our country, atractyloside, which is abundant in Xanthium strumarium L. plant, also known as pitrak, has been subject to many experimental studies since it poses a potential risk. In this context, in our current thesis, it was aimed to determine some toxicokinetic parameters of Atr given to rats orally for the first time. In our study, the seeds of spring plants collected from Bingöl and its region in autumn were used. The seeds of the Xanthium strumarium L., obtained from Bingöl and around, were removed from the prickly capsules and extracted after grinding. The amount of Atr in the extract was determined with the GC-MS device floowing hydrolysis of this solution with HCl, and mixing with ethyl acetate and derivatization step by pyridine-TMSI solution. A total of 72 Wistar Albino male rats were used in the treatments and control groups. One by one, all treatments rats were given a single dose of Atr at 80 mg / kg live weight by gavage application. Blood samples were taken from the rats in the control and experimental groups at 0., 4., 6., 8., 12., 24., 36., 48., 60., 72., 96. and 120. hours, via euthanasia using decapitation method. Atr measurements in the blood were calculated by the GC-MS device. After oral administration of Atr to the rats, absorption rate constant (ka) 0,114 h, the maximum blood concentration (Cmax) 10,77 μg / ml, the time to reach the maximum blood concentration the (tmax) was 48 h, half-life of Atr is (t1/2)) 6,071 4 h, the area under the time curve of serum drug concentration (AUC 0 → 120) 430,231 μg / hr / ml, the time required to remove 63.2 % of Atr is (MRT) 47,807 hours, distribution volume (Vdss) 1,623 ml / kg; clearance (Cl) was calculated as 0,185 ml / hr / kg, As a result, some toxicokinetic parameters obtained from our study revealed that the level of Atr in the blood was fast, reliable, simple analytical and sensitive.
