Effect ofrhIL?11 in retinal ischemia-reperfusion

Abstract (EN)

This study was performed in the Ophthalmology Department of Firat University Medical School.The purpose of this study was to determine the protective effect of recombinant human IL?11 in guinea pigretina that was exposed to ischemia?reperfusion injury.We induced anesthesia and analgesia in all guinea pigs except for the placebo group. Then retinalischemia was induced in group 2 and 3 by increasing the intraocular pressure to 150 mmHg for 90minutes.We have administered 5 ?g/kg intraperitonally recombinant IL?11 in group 3, before the onset ofischemia. After 90 minutes of ischemia, intraocular pressure decreased to normal values and reperfusionwas induced for 2 days. Interleukine?11 group was administered intraperitonally recombinant IL?11 every24 hours during the reperfusion period. After 2 days the eyes were enucleated.The mean retinal tissue values of TNF??, TGF?ß, IL?6 in the sham group were statistically higherthan those of the plasebo group (p=0.008, p=0.008, p=0.008). The mean retinal tissue values of TNF??,TGF?ß, IL?6 in rhIL?11 treated group were observed to be significantly lower than those of the sham group(p=0.008, p=0.032, p=0.008).Histopathologic analysis of each group indicated that rhIL?11 group had not significantly (p=0.095)protective effect in the inner plexiform layer of retina. But also histopathologic analysis of each groupindicated that rhIL?11 group had statistically significant (p=0.032) protective effect in the inner layersof retina (internal limitan membran and inner plexiform layers) by preventing PNL infiltration in these layers.Our results suggest that according to retinal TNF??, TGF?ß, IL?6 levels and histopathologicalchanges in eyes those were induced to ischemia?reperfusion, rhIL?11 have protective role bydecreasing retinal TNF??, TGF?ß, IL?6 levels and preventing PNL infiltration in the inner layers of retina.Key words: Recombinant IL?11, ischemia?reperfusion, retina.
