vABSTRACTTHE EFFECT OF CYCLOSPORINE-A AND DEFEROXAMINE ONEXPERIMENTAL INTRACEREBRAL HEMORRHAGE IN RATSDr.Ilhan BUYUKBESEResidency Thesis, Department of NeurosurgeryThesis moderator: Prof.Dr. Abdulvahap GOKMay 2007, 68 pagesIn thıs study the effects of immunosupressor Cyclosporine-A (Cs-A) and ironchelator deferoxamine (DFO) were investigated histopathologically on experimentalintracerebral hematomas (ICH) in rats.Thirty eight wistar rats were used. After a stereotactic frame was applied to allanimals, a burr-hole was made and intracerebral hemorrhage occured by injecting 0,2 ccautologous blood.The animals were divided in four groups. In group I (n=9) the loading dose of Cs-A(20mg/kg) and DFO (100 mg/kg) was given intraperitoneally in the first 30th minute. Ingroup II (n=9) and group III, (n=10) the same doses of the drugs were given at the 1stand 4th hours respectively. In group IV (n=10,control group) no treatment was ofapplied. Cs-A in a dosage of 10 mg/kg once a day and DFO 100 mg/kg two times a daywere given intraperitoneally for three days on a continuiting of the treatment. Theanimals were sacrificed after 3 days for histopathological evaluation.The histopathological differences of groups induced by oedema, inflamation andnecrosis were investigated by Kruskall -Wallis variant analysis and Mann Whitney-Utest. Significant statistical decrease was observed in oedema formation (p<0,05) in thetreated groups, but no changes were established in inflamation and necrosis (p>0,05).The neuroprotective mechanism of Cs-A and DFO hasn?t been known yet. It is probablethat. The mechanism could be inhibition of cell injury due to Fe++ and Ca++ ions.Key words: Deforoxamine, Intracerebral hematoma, Cyclosporine-A
