The effects of salvia officinalis in TNBS-induced experimental colitis model in rats

Abstract (EN)

Objectives: The present study was aimed at investigating the effects of Salvia officinalis on damage produced following TNBS -induced colitis in ratsDesign: 110 healthy young male albino Wistar rats were randomly divided into 11 groups: control group, colitis group (for three days), sesame oil group (for three days, SY-3), 0.5 mg/kg Salvia officinalis extract group ( for three days, 0.5 SO-3), 5mg/kg SO extract group ( for three days, 5 SO-3), 50mg/kg SO extract group ( for three days, 50 SO-3), colitis group ( for seven days), sesame oil group ( for seven days, SY-7), 0.5 mg/kg SO extract group (for seven days , 0.5 SO-7), 5mg/kg SO extract group (for seven days , 5 SO-7), 50 mg/kg SO extract group (for seven days, 50 SO-7). The rats were starved 12 hours before the induction of colitis. polyethylene catheters were inserted through the rectum into the colon, under ketamine (50mg/kg) and Xylazine (5mg/kg) anaesthesia , with the bowel being empty. Injecting via a catheter 0.8 ml of 25 mg of TBNS solution dissolved in 37% ethanol, colitis was induced. On the third and seventh days of the induction, the rats were sacrified and the samples of the colonic tissues were obtained. The levels of malonyl-dialdehyde (MDA), and the activities of superoxide dismutase (SOD), catalase (CAT), nitric oxide (NO), myeloperoxidase (MPO), glutathione reductase (GR), and glutathione peroxidase (GSH-Px) were measured in colonic tissues.Results: Macroscopic improvement occurred on the seventh day in those groups receiving liquid extract of 0.5 mg/kg and 50 mg/kg Salvia officinalis ( p< 0.05). On the third day, the levels of NO were found to be significantly lower in the groups receiving liquid extract of 0.5mg/kg and 5mg/kg Salvia officinalis than those in the colitis control groups (p< 0.05). On the third day, SOD levels were found to be significantly higher in the group receiving 50mg/kg Salvia officinalis liquid extract than in the colitis control group (p<0.05).Conclusion: In the present study, it was observed that Salvia officinalis-treated groups displayed a considerable improvement in colitis It was also seen that Salvia officinalis has beneficial effects on the course of inlammatory bowel disease, including ulcerative colitis and Crohn disease which are likely to operate through anti-inflammatory and anti-oxidant mechanisms. Therefore, Salvia officinalis should be considered a useful addition to the treatment of colitis.
