Abstract (EN)

In this study, the effect of SPCC320.03 gene (which is the orthologist of the RGT1 gene encoding the Rgt1 transcription factor in Saccharomyces cerevisiae) on the transcription of the hexose transporters in Schizosaccharomyces pombe strains produced in different carbon sources was investigated. The effects of different sources on the transcription of the scr1 gene, which is a transcription factor in the carbon catabolite mechanism, and the fbp1 gene, which is involved in the gluconeogenesis pathway, was performed as a control of the efficiency of the experiment. For this purpose, the SPCC320.03 gene was ligated with the pSLF272 vector and transformed into two S. pombe strains, EDD666 and SPCC320.03Δ. The transformation proved successful by PCR and western blot analysis, followed by RNA expression levels of the relevant genes in S. pombe strains grown in medium containing 2% glucose, 2% gluconate and 2% glycerol + 0.1% glucose by Real Time PCR method. Compared to EDD666 gene expression rate, in SPCC320.03Δ gene expression rate; The gene expression levels of the ght3, ght4 and ght6 genes increased with 2% gluconate and 2% glycerol + 0.1% glucose; It was observed that expression of ght2 gene decreased in 2% glycerol + 0.1% glucose environment. With the results obtained, strong evidence is presented that the SPCC320.03 gene can be a transcription factor that can negatively regulate the expression of the ght3, ght4 and ght6 glucose transporter genes in glucose starvation and positively regulate the expression of the ght2 glucose transporter gene.
