Abstract (EN)

Mankind has been asking the question "Why do we age?"and tried to find answers to this question for centuries. Today, the causes of aging are still being researched and many new theories about the causes of aging are produced. Knowing the factors that affect aging and taking some precautions about them will lead to an important step to be taken in order to lead a healthier and longer life, even if aging cannot be completely prevented. Metformin (N,N-dimethylbiguanide) is the most commonly used active ingredient in the treatment of type-2 diabetes due to its potential to lower blood glucose. In this widespread use process, it has been shown that metformin has anti-aging properties and longevity in different organisms, as well as its protective properties against heart diseases and cancer. It also has anti-aging properties not only through its effect on glucose metabolism, but also through decreasing Insulin-Like Growth Factor-1 (IGF-1) signal transduction, inhibiting mTOR signalling pathway, activating AMPK signalling pathway, suppressing mitochondrial complex 1 in the electron transport system, and reducing reactive oxygen species. In this study, the effect of metformin on cellular aging was investigated by using wild strain (972h-) of Schizosaccharomyces pombe, a widely used model organism. For this purpose, after different doses of metformin were applied to the cells, the effective dose was determined and the effect of this effective dose on some processes such as chronological lifespan, glucose xv consumption, ATP production, amount of oxidative stress and resistance to environmental stress was examined. When the results were examined, it was seen that metformin increased the lifespan of S. pombe with its positive effects on changes in the aging process.
