Abstract (EN)
Diabetes mellitus is a common metabolic disease and its incidence increases dramatically. With the spread of diabetes mellitus, there is a significant increase in the complications of disease. Diabetic neuropathy is the most common disease among these complications. Therefore, candidate natural agents have been searching for various medicines for relieving, protecting or remediation against diabetic neuropathy. Flavonoids, which are phenolic compounds found in plants, have antidiabetic and antioxidant effects. Neoeriocitrin is a newly discovered flavonoid and there is not much information about its effect. Therefore, this study has been planned by considering antioxidant effects of flavonoids, neoeriocitrin may be a candidate agent. In this study, the protective effect of neoeriocrine was investigated by causing damage to SH-SY5Y neuroblastoma cells with high glucose concentration. In the study, crystal violet and trypan blue staining were performed to determine cell viability. In order to determine its antioxidant properties, the amount of intracellular ROS resulting from oxidative stress and the amount of MDA, iv which is a lipid peroxidation marker, were measured. According to the results obtained, it was found that neoeriocitrin decreased the amount of ROS at increasing concentrations and had an effect on the reduction of lipid peroxidation at a concentration of 20 µM. On the other hand, ATP levels were measured to determine the metabolic activity of the cell and apoptotic cell death determined by the TUNEL test. According to the results, it was found that the amount of ATP increased and the number of apoptotic cells decreased in SH-SY5Y neuroblastoma cells at increasing concentrations of neoeriocitrin. Considering all the results obtained from the study, it was revealed that neoeriocitrin had a protective effect against the damage in SH-SY5Y cells that were modeled for diabetic neuropathy with high glucose concentration
