Abstract (EN)

In this thesis, it is aimed to determine the most suitable conditions for the in vitro propagation of the endemic and endangered Stachys cretica L. subsp. kutahyensis Akçiçek subspecies, which has medicinal and economic importance. Diferent concentrations of Plant Preservative Mixture, Gibberellic Acid ve Juglone were used for in vitro germination. Seedlings, obtained from in vitro germinated plantlet, were used as an explant sources in tissue culture studies. It was determined that the best shoot regeneration in Stachys cretica L. subsp. kutahyensis Akçiçek subspecies was achieved in 2 mg/L BAP treatment. The effects of MS and different concentrations of NAA, IBA, and juglone as well as growth regulators such as Mod MS and different concentrations of juglone on the number of roots per explant, root length, and root growth percentage were investigated on rooting development. It was observed that the most successful medium in root formation was Mod MS medium containing 10-7 M juglone. This was followed by Mod MS containing 10-6 M juglone. Rooted plantlets were gradually transferred to the soil by adapted external conditions. At the end of the two-week period, all of the seedlings survived. The transfer of Stachys cretica L. subsp. kutahyensis Akçiçek plants, which were adapted to the soil, to the field conditions (Tavşanlı/Nusretler) was successfully carried out.
