Purification of invertase enzyme from Stevia rebaudiana and Momordica charantia by TPP method

Abstract (EN)
This study was aimed to purify a widespread used enzyme particularly in the food industry intervase from an unused phytotherapy plant Momordica charantia via developing a single step TPP method and to identify the characterization of the pure enzyme. Optimum conditions for TPP method was determined for the purification of Invertase, as a result, 70% ammonium sulfate saturation, 1: 0.5 t-butanol ratio, and a 30 minute incubation time lead to 20.28% efficiency and 10.48 fold partially purification. After TPP, total protein was decreased from 34.86 mg to 0.675 mg in the homogenate, and the specific activity value was increased from 21.672 U/ mg protein to 227.022 U/ mg protein. In conclusion, the optimized single step TPP method revealed a better purification comparing to several purification methods in the literature.
