Determination of ability of alpha amylase production by Streptomyces sp. MC10 strain

Abstract (EN)

Amylases are one of the most important and industrial enzymes. In this study, the amylase from Streptomyces MC10 strain isolated from soil was produced and characterized. The strain was tested for determination of temperature, pH range and NaCI concentration for growth and enzyme production on plate. It was found the Streptomyces MC10 had the best enzyme activity according to raw starch degrading amylase screening methods, and Streptomyces MC10 was selected as amylase source. Arabinose, glucose, mannitol, sucrose and soluble starch were tested to determine the best carbon source. Soluble starch was determined as the best carbon source. Peptone, (NH4)2SO4, L-asparagine, urea and KNO3 were tested to determine the best nitrogen source. Peptone was determined as the best nitrogen source. DNS method was used in determining the amount of amylases. The optimum fermentation conditions were found to be incubation period of 4 days, inoculum concentration of %1, NaCI concentration of 5 gr and incubation temperature at 28°C pH 7. This amylase showed maximum activity at 50°C and pH 7. Key Words: ?-amylase, Streptomyces, soluble starch
