Abstract (EN)

In this thesis study, the in vitro micropropagation method of Oxalis triangularis A.St.-Hil, a medicinal and ornamental plant that draws attention with its purple leaves and unique phototropic behavior, was investigated. Leaf and petiole explants obtained from healthy plants grown in laboratory environment were used as thesis material. As a result of many sterilization experiments, 100% sterile and 80% healthy explants were obtained in soaking them in 70% ethanol for 1 min and 0.5% NaOCl for 1 min. Sterile explants were transferred to Murashige and Skoog (MS) media containing benzyl amino purine and naphthalene acetic acid plant growth regulators in different concentrations and combinations, namely; M1 (0.5 mg/L BAP), M2 (0.5 mg/L BAP + 0.5 mg/L NAA), M3 (0.5 mg/L BAP + 1 mg/L NAA), M4 (0.5 mg/L BAP + 2 mg/L NAA), M5 (1 mg/L BAP) in a sterile cabinet. Culture procedures were carried out in plant growth chamber with a temperature of 24±2°C, a light intensity of 4000 lux and a 16-hour light/8-hour dark cycle. Morphological changes in the explants were observed and the obtained data were evaluated using the Minitab statistical analysis program. The best results were obtained in leaf explants with 0.5 mg/L BAP and 2 mg/L NAA application. According to statistical analysis, the average number of shoots per explant was determined as 5.67 a ±2.43 and the number of leaves per explant was determined as 4.00 a ±1.86. The obtained shoots were rooted in MS nutrient medium containing 0.5 mg/L NAA. The data obtained as a result of this study, which investigated the in vitro production of Oxalis triangularis A.St.-Hil, which is medically and economically valuable, is of a guiding nature for many studies such as basic sciences, genetics and micropropagation that can be done about the plant.
