Abstract (EN)

11 ABSTRACT Ph.D Thesis PREPARATION OF HYDROPHOBIC ADSORBENT BASED ON TETRAPHENYLPORPHYRIN TURGAY TAY Anadolu University Graduate School of Sciences Chemistry Program Superviser: Assoc.Prof.Dr. Hayrettin TURK 2004, 108 Pages In this study, a polymeric adsorbent with tetraphenylporphyrin Iigand was prepared as an hydrophobic interaction adsorbent and the adsorption behaviour of bovine serum albumin (BSA) on this adsorbent was studies. As a polymerizable porphyrin, 5-(4-metacryIamidophenyl)-10,15,20-triphenylporphyrin was synthesized in three steps. The polymeric adsorbent with tetraphenylporphyrin Iigand was prepared from monomers 5-(4-metacrylamidophenyl)-10,15,20-triphenyIporphyrin (1%), acrylamide (39%), and cross-linking agent ethyleneglycoldimetacrylate (60%) using suspension polymerization technique. The adsorption behavior of bovine serum albumin on this adsorbent was studied as a function of the initial BSA concentration, pH, ionic strength, and type of salt Very high adsorption capacity (qm) and low dissociation constant (Ka) for the adsorbent were observed i.e. 775 mg BSA/g adsorbent and 1.21xl0"2 mg/mL at pH 4.0 and 625 mg BSA/g-adsorbent and 0.170 mg/mL at pH 7.0 in 1.0 M Na2S04 and buffered solutions. In addition the amount of adsorbed BSA increased as the ionic strength of the solution increased. The adsorbent showed hydrophobic interaction adsorbent behavior and van der Waals forces and n-n interactions between BSA and PFA were the major factors for high adsorption capacity. The adsorption process was nearly irreversible on this adsorbent. Keywords: Adsorption, Bovine Serum Albumin, BSA, Porphyrin, Hydrophobic Interaction
