In this study, the antiproliferative effect of Thymus vulgaris L. plant was studied on oral cancer cells and its cytotoxic effect was evaluated. According to these results, 1:10, 70% ethanol extract was found to be quite effective on oral cancer cells. Within the scope of the thesis, dried Thymus vulgaris plant's herba 70% ethanol extract (1:10) and aqueous (1:10) extract were prepared and the cytotoxicity of these extracts on OSC-19 and HBT-41 cell lines was studied and it was determined that 70% ethanol extract exhibited cytotoxicity. T.vulgaris herba standardized 70% ethanol extract and aqueous extract were obtained. The specifications of each were established. According to the experimental results, 15 µg/ml dose of Thymus vulgaris extract prepared in alcohol, proliferation was 21% for OSC-19 cells; It was determined that HTB-41 suppressed by 7.4%. Thymus vulgaris extract prepared in water was not found to be effective for both cells. This study, the antiproliferative and cytotoxic effects of Thymus vulgaris L. (common thyme) on oral cancer cell lines were evaluated. The aim was to investigate the potential therapeutic effects of different extracts of the plant on oral cancer cell lines OSC-19 and HTB-41. Two types of extracts were prepared from the dried aerial parts of Thymus vulgaris : a 70% ethanol extract and an aqueous extract, both at a 1:10 (w/v) ratio. These extracts were applied to cultured cancer cells and the effects on cell viability and proliferation were examined. The cytotoxicity analyses revealed that the 70% ethanol extract exhibited a significant antiproliferative effect on both cell lines, whereas the aqueous extract showed no notable cytotoxic or antiproliferative activity.According to the results, the ethanol-based extract at a concentration of 15 µg/ml inhibited proliferation by 21% in OSC-19 cells and 7.4% in HTB-41 cells. This indicates that the active compounds responsible for the biological activity are more effectively extracted in ethanol, highlighting the importance of the solvent used during extraction. Additionally, both extracts were standardized and their specifications were established. In conclusion, the ethanol extract of Thymus vulgaris demonstrated a clear anticancer potential, particularly in the OSC-19 oral cancer cell line. These findings provide preliminary insights for future pharmacological and clinical studies, supporting the possible role of natural products in cancer therapy.
