YFHA-YFHK and CUSS-CUSR two-component systems stress different conditions (pH, osmolarity, some antibiotics and metals) investigation of Escherichia coli W3110 life in roles

Abstract (EN)

Bacteria have a signal transduction called a two-component system in order to adapt to external environmental stimuli. From these systems, some functions of CusS-CusR and YfhK-YfhA two-component phosphorylation systems have been identified. However, it has been thought that these systems have different roles under different stress conditions other than designated ones. Therefore as a first step, mutants of these systems were obtained and their roles under different conditions were investigated. Their roles were investigated against pH (pH 5.5, 7.0, and 8.5), osmolarity (0.2 M, 0.1 M, 0.01M), various metals (Cu, Co, Zn) and antibiotics (Kanamycin, Tetracycline, Streptomycin) effect. In the study, mutants obtained from abroad were transferred to the bacteria Escherichia coli W3110 through P1 transduction method. In addition, by means of the plasmid obtained from abroad, the roles of the genes in the studied stress conditions were controlled through performing by complementation tests. At the end of the study, obtained data were analyzed statistically through student t test. As a result of this analysis, it was found that studied genes had no roles in pH 7 which was neutral pH. yfhA, cusS and cusR genes which was studied for life under alkaline pH conditions had important roles. yfhK, cusS and cusR had importance in acidic pH conditions. Under osmotic stres conditions, yfhA yfhK, cusS and cusR had important roles (p˂0.05). While yfhA, yfhK, cusS and cusR for Zn and other genes except yfhA and cusR against Co metals had important roles, 3 genes except cusS against Cu stres had roles. Genes which was determined unimportant to protect against metal stress, was found insignificant according to statistically analysis. All genes especially against tetrasiklin 3 genes except cusS had roles. 3 genes, except yfhK, had important roles against streptomisin, and it was found that resistance occured as a result of genes deletion. It has been recommend that it should be studied in the next projects whether studied genes controls the directly related genes with stress factors.
