Effects of 5-fluorouracil and proteasome inhibitor bortezomib on 4T1 breast cancer cells

Abstract (EN)

In this study, the effects of 5-fluorouracil, proteasome inhibitor bortezomib, which one used in cancer therapy, and the combinations of 5-fluorouracil + bortezomib 4T1 in breast cancer cells are examined. In our study, 4T1 cells cancer cells were treated with for 24 hours with different concentrations of 5-fluorouracil, then MTT assay was used to determine % survival and IC50 values were calculated. The results showed that the IC50 value of 5-fluorouracil was 8.9 ?M in 4T1 cells. Afterwards, in 4T1 cancer cells % survival of control (DMSO), 5-fluorouracil, bortezomib and that of the combinations of 5-fluorouracil + bortezomib at 24 h was compared with each other. In addition, the effects of combinations of these drugs are also examined by soft agar colony formation assay in 4T1 cells. Apart from these, the combination of 5-fluorouracil and bortezomib was investigated to determine whether a new treatment can be developed a for breast cancer. In light of these results, it is thought that a new treatment method would be developed if 5-fluorouracil and proteosome inhibitor bortezomib combination is tested in clinical studies.
