The investigation of the effects of adrenomedullin and methyl adrenomedullin treatment on some endogen angiogenic and biochemical factors

Abstract (EN)

Adrenomedullin is a potent vasodilator and hipotensive peptid that was originally isolated from human pheochromocytoma. There is a lot of evidence that adrenomedullin plays important roles in many biological functions as a otocrine, paracrine and endokrin mediators. Recently, it has reported that Adrenomedullin has an angiogenic property. Most proteins undergo post-translational modifications. These modifications like methylation, phosphorylation are extremely important because they provide important biological molecules become functional.In this study, we investigated the effects of adrenomedullin and methylated adrenomedullin treatment on some endogen angiogenic factors and biochemical changes. Thirty-six male wistar rats were divided into six groups: Control group, Stress group, Adrenomedullin group, methylated adrenomedullin group, stress+adrenomedullin group, stress+ methylated adrenomedullin group. In AdM treated group and met-AdM treated groups, animals received intraperitoneal (i.p) injection of AdM and met-AdM (2000ng/kg body weight) once a day during a week. Stress group animals were exposed +10 °C cold during a week. The activities of catalase (CAT), superoxide dismutase (SOD), glutathione peroxidase (GSH-PX) and matrix metalloproteinase (MMP-2) enzymes activities and nitric okside (NO), vascular endothelial growth factor (VEGF), tumor necrosis factor-? (TNF- ?) and interleukin-6 (IL-6) levels were determined in the tissues of rats. Total RNA levels were determined in liver, lung, kidney and heart tissue of rats.In conclusion, the groups treated AdM with stress, there was a compensating effect of AdM on some antioxidant enzymes in all tissues. The groups treated met-AdM with stress, there was a compensating effect of met-AdM on some antioxidant enzymes in all tissues except heart. The AdM and stress application, AdM increases VEGF and NO levels. However, it was determined that effects of AdM on IL-6 and TNF-? levels and MMP-2 activity showed differences in various tissues. Stress+ Met AdM treatment effected NO and VEGF levels paralel with AdM, conclisions but IL-6, TNF-? levels and MMP-2 activity showed differences in various tissues.
