Abstract (EN)
In this study, leaves of wild pear (Pyrus elaeagnifolia) which have local names such as 'cördük' belonging to flora of Afyonkarahisar Büyükkalecik were used. The leaves of wild pear were collected from the mountains and dried for about 2 weeks before being exposed to direct sunlight. Extraction step was applied by grinding of dried leaves. Extraction time (30-45-60-75-80 min), temperature (25-30-40-55-70oC), ethanol con-centration (% 0-20-40-60-80) were studied by varying the parameters. The characteriza-tion of the phenolics in the leaf composition was carried out by HPLC. Chlorogenic acid (2,2%) has the highest concentration in the leaf, this was followed by catechin hydrate (1,1%) and routine (0,4%). Total phenolic content of samples was determined by Folin - Ciocalteu method. The highest values in the parameters were observed at approximately 45 minutes at 40 °C and % 60 concentration. DPPH method was used to determine the antioxidant capacity of samples. Antioxidant capacity showed an increase until 75 mi-nutes, 40 °C and %60 concentration, then a decrease was observed. The antimicrobial activities of extracts on the microorganisms such as Staphylococus aureus, Bacillus sub-tilis, Bacillus cereus, Escherichia coli, Listeria monocytogenes and Pseudomonas aeru-ginosa were determined by disk diffusion method. The cytotoxic effect of extracts on MCF-7 ile A549 of cancer cells was determined by MTT (3-(4,5)-dimethylthiazol-2yl)-2,5-dipheniyl tetrazolium bromide) test at optimum extraction conditions. However, it has been found that A549 also has a significant effect at low concentrations in cancer cells.
