Determination of the anti-cancer effects of agomelatine

Cancer is a disease that is increasingly common in the world, ranking second after cardiovascular diseases. Ovarian cancer is the type of cancer with the highest mortality rate among gynecologic cancers. Agomelatine is a synthetic analogue of the melatonin hormone developed as an antidepressant drug. Studies investigating its antidepressant effects are on the rise. Cisplatin is a potent anticancer agent. It is widely used alone and in combination in cancer treatments. miR-340-5p is highly conserved across mammals and plays a crucial role in cancer pathogenesis. In this thesis, miR-340-5p transfection procedures were performed after determining the IC50 doses of agomelatine and cisplatin with the appropriate combination doses on SKOV-3 ovarian cancer cells. Agomelatine IC50, cisplatin IC50, and combined 40% doses were applied to the miR-340-5p transfected SKOV-3 cells, and their effects on cell proliferation and migration were investigated by the real-time analysis (RTCA) method. In the experimental results obtained, it was determined that the miR-340-5p transfected SKOV-3 cell lines, a proliferation rate in agomelatine IC50, cisplatin IC50, and combined 40% drug-treated cell groups, while the migration rate decreased in the cell groups treated with agomelatine IC50. Apoptosis cell ratios were determined by the Annexin V-FITC kit and Acridine orange/Ethidium bromide staining method, and the apoptotic cell number increased in SKOV-3 cell lines transfected with miR-340-5p after application of the drug dose. The results obtained show that agomelatine may be effective in anti-cancer and that combining treatment may be effective in combination rather than using them separately in treatment.
