Isolation and characterization of various bacteria in some regional yoghurt samples in Agri

Abstract (EN)

In this study, various Bacteria of some local yoghurt samples in Ağrı province have been isolated and cultured and characterized. In order to represent the Ağrı province, 11 yoghurt samples were taken from 5 different villages and 28 isolates were obtained from these yoghurt samples. In the first step, morphological tests (Gram, mobility and morphology), the physiological tests (salt, pH and temperature tolerances), biochemical tests (oxidase, catalase, utilization of arginine and esculine, gas production from glucose) and antibiotic susceptibility (neomycin and tetracycline) of these 28 isolates have been performed. Within the molecular tests scope, DNA extraction and pcr amplification of 16S rDNA region of these 28 isolates have been performed. Among these 28 isolates, the 16S rDNA sequence of 24 isolates has been obtained and the rest 4 isolates have been characterised with WITEK 2 method, yet, no identification data was obtained. According to nucleotide blast data obtained from NCBI, the similarity of the isolates ranged between 99 and 89 %. 99% similarities were obtained from 4 isolates of Sphingonomas sp and each of the Lactococcus lactis subsp. lactis, Lactobacillus casei ve Lactobacillus paracasei strains.
