Investigation of miR-423-5p and miR-664b-5p in patients with familial and sporadic ovarian cancer

Abstract (EN)
MicroRNAs(miRNAs) are endogenous,non-coding,single-stranded RNA molecules about 22 nucleotides in length.Ovarian cancer is a heterogeneous disease with different biological behaviors at clinical and molecular levels.miRNAs are overexpressed and play roles in the regulation of mammalian growth,early diagnosis of ovarian cancer,prognosis and chemotherapy sensitivity.In our study,the expression levels of miR-423-5p and miR-664b-5p,which are selected from the earlier studies and found to be important for the etiology of ovarian cancer,were investigated in the peripheral blood of familial and sporadic ovarian cancer patients and healthy individuals.Expression analyzes by using Real Time PCR technique,were evaluated statistically with SPSS v21.0 program.According to these results;there was a statistically significant difference between healthy controls and ovarian cancer groups for both miRNAs(p=0,000).In patients with ovarian cancer,the expression of miR-423-5p increased by 2.35 times and the expression of miR-664b-5p increased by 2.47 times compared to the control group.In the literature,it has been reported that miR-423-5p has been shown to be overexpressed in many cancer types and different disease stages except ovarian cancer.In our thesis,it has been shown for the 1st time in the peripheral circulating lymphocytes of ovarian cancer cases.Generally,in the group of ovarian cancer miR-664b-5p expression level was increased but it was found to be decreased in 60% of BRCA mutation carriers among cases with ovarian cancer. In addition,it is understood that these miRNA molecules have the potential to be a non-invasive biomarker for ovarian cancer and it is useful to show the changes of these molecules in future studies,on tumor tissue and during treatment.
