The effect of carbolatin ve curcumin combinations onoxidative stress in lung cancer

Abstract (EN)

Lung cancer is one of the the primary causes of cancer related deaths both in men and women. Carboplatin (CARB) which is one of the antineoplastic agents of the platinum group is efficiently used as a chemotherapy agent in a various of cancer types such as ovary, lung, and brain cancer. Curcumin (CUR) is a natural polyphenol compound which has antioxidant, antiinflammatory and antiproliferative features. In this dissertation effects of use of CARB and CUR a combination to cell lines in CCD-19Lu lungs fibroblast and oxidative stress in the A549 lung adenocarcinoma were examined. The cytotoxic effects of CARB and CUR's were determined by using MTT method. IC50 values of CARB and CUR in the CCD-19Lu were calculated respectively 200±16 µM; 150±12 µM and 100±9 µM; 60±8 µM in A549 cell lines. While determining oxidative stress, Oxidative Stress Index (OSI) was calculated by measurement of Total Oxidant (TOS) / Total Anti-Oxydant (TAS) phases. In both cell lines it is monitored that single use of CARB increased the OSI value while combination generated with CUR decreased the OSI value. Other markers of oxydative stress; such as superoxide dismutase (SOD), reduced glutathione (GSH), and malondialdehyde (MDA) levels were also analysed. It was determined that combination groups generated with CUR in the A549 cell line (KM30; KM40), SOD levels was monitored (1,086±0,09 U/mL) respectively as 0,891±0,02 U/mL; and GSH levels was monitored (0,3590±0,02 µM) respectively as 0,3870±0,03 µM; 0,3905±0,02 µM, and MDA levels was monitored (1,050±0,05 µM) respectively 0,740±0,04 µM; 0,622±0,02 µM.
