Abstract (EN)
Aim. Objective of the present study is to develop UV-VIS Spectrophotometry and LC-MS/MS methods and perform their validity tests for the determination of Albendazole (ABZ) and its metabolites [albendazole sulphoxide (ABZSO) and albendazole sulphon (ABZSO2)]. Develop LC-MS/MS method in human and rabbit plasma by performing its validity tests. In order to show the applicability of the methods developed, quantity of ABZ, ABZSO and ABZSO2 was in both pharmaceutics preparats and in the plasma of rabbit given albendazole orally determined. In addition, pharmacokinetic parameters were determined. Material and Method. UV-VIS Spectrophotometry was performed at 295.0 nm wavelength. In LC-MS/MS method, study parameters, C18 column, isocratic elution and methanol:water were used. Results. UV-VIS spectroscopy method was linear in 2-20 µg/mL concentration range, its relative error was lower than %7 and Relative Standard Deviation of the method is better than %0.9 while Mean Analytical Recovery value was %100.5. In standard solutions, human plasma and rabbit plasma studies, linearity range was 5-1000 ng/mL, %BH was lower than %6.0 and %RSD was better than %0.9. Mean Analytical recovery was measured as %100 and %99.5 for pharmaceutical formulations and human plasma, respectively. Concentration-time graphs were derivated for oral administration of Andazol tablet and Andazol Suspension and pharmacokinetic parameters were determined. Conclusion. The applicability of the developed and validated method was monitored in both pharmaceutical formulations and plasma samples LC-MS/MS and UV-Vis spectrsocopy methods by this proposed study.
