Investigation of the protective role of resveratrol against aflatoxin B2 toxicity in albino mice

Abstract (EN)

Physiological, biochemical and cytogenetic parameters were used to determine the toxic effects. For this purpose, 6 experimental groups, including 6 albino mice in each group, were formed. In experimental period, group I was fed with pellet feed and water as the control group. 10 mg/kg resveratrol, 20 mg/kg resveratrol, 20 μg/kg aflatoxin B2, 10 mg/kg resveratrol + 20 μg/kg aflatoxin B2, 20 mg/kg resveratrol + 20 μg/kg aflatoxin B2 were administered to the group II, III, IV, V and VI, respectively. Physiological changes were investigated by measuring the body weight, liver and kidney weight of the mice before and after the application. Biochemical parameters were investigated by measuring alanine transaminase, aspartate transaminase, blood urea nitrogen and creatinine levels in serum samples, malondialdehyde and glutathione levels in kidney and liver tissues. Genotoxic effects were investigated by determining the frequency of micronucleus in buccal epithelium, erythrocyte and leukocyte cells, and the frequency of chromosomal abnormalities in bone marrow cells. As a result, while significant decreases were detected in body weight, kidney and liver weights in the aflatoxin B2 applied group, it was determined that these decreases regressed in the aflatoxin B2 + resveratrol groups. It was found that the serum alanine transaminase, aspartate transaminase, blood urea nitrogen and creatinine parameters levels of the aflatoxin B2 treated group were higher than the control group, and there were serious abnormalities in the levels of malondialdehyde and glutathione in the kidney and liver. These results indicate that aflatoxin B2 administration causes liver and kidney damage and this damage is caused by oxidative stress. Significant improvements in biochemical parameters were observed in aflatoxin B2+20 mg/kg resveratrol treated groups. It has been determined that aflatoxin B2 induces micronucleus formation in buccal epithelium, erythrocyte and leukocyte cells, and chromosomal damage such as fracture, fragment, ascentric chromosome, dicentric chromosome, gap and ring in bone marrow cells. It has been observed that administration of 20 mg/kg resveratrol together with aflatoxin B2 decreases the frequency of micronucleus and chromosomal abnormalities. In this study it was determined that aflatoxin B2, a mycotoxin, caused serious changes in selected biochemical and physiological parameters and genotoxic effect in albino mice, while resveratrol has a protective role against these toxic effects with its antioxidant properties.
