Determination of genetic diversity of brown layer pure lines in the Ankara Poultry Research Station by using microsatellite markes

Abstract (EN)

In this study, genetic diversity was investigated in different six brown layer lines (RIRI, RIRII, BARI, BARII, L-54, COL) in the Ankara Poultry Research Station. A total of 180 chickens belonging to six lines RIRI (n=30), RIRII (n=30), BARI (n=30), BARII (n=30), L-54 (n=30), COL (n=30) were genotyped using 22 microsatellite markers. Mean number of alleles per locus (Na), effective number of alleles (Ne), polymorphic information content (PIC) were calculated at RIRI line 5.45, 3.11, 0.58; RIRII line 4.95, 2.44, 0.49; BARI line 5.00, 2.85, 0.55; BARII line 4.41, 2.62, 0.53; L-54 line 4.86, 2.64, 0,51 and COL line 4.73, 2.47, 0.50 respectively. Observed heterozygosity (Ho), expected heterozygosity (He) and inbreeding coefficient (Fis) values were detected at RIRI line 0.48, 0.64, 0.28; RIRII line 0.31, 0.54, 0.46; BARI line 0.44, 0.61, 0.27, BARII line 0.50, 0.59, 0.18; L-54 line 0.47, 0.56, 0.16 and COL line 0.37, 0.56, 0.35, respectively. F-statistics (FIS, FIT, FST) were determined as 0.273, 0.493, 0.295, respectively across six chicken lines. Across lines mean pairwise FST value was 0.294 (p < 0.01), and coefficient of gene differentiation (GST) was found to be 0.274. According to results of Analyses of Molecular Variance (AMOVA), the total genetic variation arises from 29.5% differences among population and 17.09 % being within individuals among population. According to NJ tree, Factorial Correspondence Analysis (FCA) and Structure analysis, lines were clustered in accordance with genetic origins and breeding history. As a result, findings obtained in this study have showed that populations had intermediate levels of polymorphism and high levels of inbreeding. Genetic differentiation was also determined to be high levels between populations. In terms of sustainability of the studies at Ankara Poultry Research Station would be useful to take various measures, particularly to reduce inbreeding level in populations.
