Characterization of a novel small and circular plasmid (pAnox1)isolated from Anoxybacillus gonensis 05s15

Abstract (EN)

In this study, a small, novel, cryptic, double stranded plasmid which has 1592 bp in long and with a total G + C content of 40,01% from Anoxybacillus gonensis 05S15 was characterized. According to BLAST similarity searches performed with the nucleotide sequence of pANox1, it was found that it did not show significant similarity to other plasmids, but it had similar regions to replication proteins responsible for replication. Five ORF regions were predictied on the plasmid genome by bioinformatic analysis. Three of these ORFs are on one strand and the other two are on the opposite strand and three palindromes and two iterons were detected in both strands. The plasmid was determined to have the RC replication mode by performing a Southern hybridization study. As a result of the study, it was concluded that the pAnox1 plasmid encodes 3 transcripts and accordingly ORF-1 and ORF-3 in the same orientation and ORF-5 in the opposite orientation encodes the transcript. Based on RACE and LACE analyzes, it was revealed that the ORF1 transcript encoded by pAnox1 encoded 903 bp, the ORF3 transcript 499 bp, and the ORF5 transcript 211 bp.
