Effect of ethyl pyruvate with anti-oxidant, anti-inflammatory andanti-coagulant properties on intestinal ischemia reperfusion injury in rats

Abstract (EN)

In the study, we determined the availability of ethyl pyruvate (anti-oxidant) in order to determine a suitable marker in the early diagnosis of intestinal damage by creating an intestinal ischemia reperfusion model in rats, to prevent intestinal damage and to prevent the developing systemic inflammatory response. It was aimed to investigate the anti- inflammatory, anti-coagulant activity). The study, which was carried out in a randomized controlled experimental design between May 2023 and May 2024, was carried out in Eskişehir Osmangazi University Faculty of Medicine Department of General Surgery in Eskişehir, Turkey. The study was approved by Eskişehir Osmangazi University Faculty of Medicine Animal Experiments Local Ethics Committee (HADYEK) (Date: 23.08.2021 decision no: 857). The study was conducted on 4 groups of 8 rats, each randomly selected. Control-only laparotomy group, I/R-only temporary mesenteric ischemia group, I/R + 50 mg/kg Ethyl Pyruvate group, I/R + 100 mg/kg Ethyl Pyruvate group. A total of 32 rats were used. . Transient ischemia time was determined as 45 minutes and reperfusion time was determined as 90 minutes. After reperfusion, blood samples were taken from the inferior vena cava and tissue samples from the terminal ileum. ALT, AST, CK, LDH, BUN, Creatinine, MDA, 4HNE, TNF-α, Interleukin-1β and Interleukin-6, PT, aPTT, INR, Platelet tests were studied and tissue samples were also taken for histopathological evaluation. As a result, 100 mg/kg ethyl pyruvate showed antioxidant, anti-inflammatory, anticoagulant and antithrombotic effects in intestinal ischemia reperfusion injury. It was observed that TNF-alpha, IL-6, IL-1B, AST, ALT, BUN, Creatinine parameters were suitable biochemical markers in the early diagnosis of intestinal damage.
