Design, synthesis and investigation of biological activities of new imidazol derivatives with antifungal effect

There are many different types of fungi in nature, and some types of fungi cause various infections. Fungal infections are known as an important type of infection leading to the death of many people in the world in a year. New imidazole derivative antifungal compounds that will provide antifungal effect, which is the subject of the thesis, were synthesized and the biological activities of the synthesized compounds were tested. In biological activity studies, compounds 5d, 5e, 5f and 5h were found to be active compared to ketoconazole and fluconazole. It was observed that the compound with the same MIC50 value as ketoconazole 0.98 µg/mL, was 5d against some fungal species, and the compound with the same MIC50 (µg/mL) values with fluconazole was 5e. In this study; 8 new imidazole derivative compounds were synthesized. Analysis of the obtained compounds; 1H-NMR, 13C-NMR and HRMS spectroscopic methods are illuminated by the analysis findings. The inhibition effects of the obtained compounds on the 14α-demethylase enzyme were also investigated. Within the scope of the thesis, the structure of 14α-demethylase enzyme active sites has been elucidated by molecular modeling studies. As a result of this study, the compounds with the highest inhibition activity on 14α-demethylase enzyme were found as 5d, 5e and 5h. For the selected compounds, docking studies were performed and the enzyme active site and binding points were determined. In molecular dynamics studies, 5d, 5e and 5h compounds were found to have stable structures.
