Investigation of inhibition effects of some sulfanilamid ederivates on acetylcholinesterase and butyrylcholinesterase enzyme

Abstract (EN)

In this thesis, it is aimed to determine the effects of kinetic parameters on enzyme activities of some derivaties of sulfanilamides as an alternative to the takrin drug used in the treatment of Alzheimer's disease on acetylcholinesterase (AChE; E.C.3.1.1.7) and butyrylcholinesterase (BChE; E.C.3.1.1.8) activities and butyrylcholinesterase enzyme.The inhibitory effects of some aromatic compounds such as Sulfadiazine, Sulfamerazine, Sulfamethazine, Sulfacetamide and Sulfisoxazole on acetylcholinesterase and Butyrylcholinesterase enzyme activities were investigated. IC50 values for Sulfadiazine, Sulfamerazine, Sulfamethazine, Sulfacetamide and Sulfisoxazole that show in vitro inhibition effect for AChE were determined as 0,300 mM, 0,363 mM, 0,353 mM, 0,507 mM and 0,0049 mM respectively. Respectively, standard deviations and mean Ki constants for the same samples were calculated as 118±0,024 mM, 0,717±0,193 mM, 0,302±0,116 mM, 0,343±0,055 mM, 0,0049±0,00012 m. IC50 values for Sulfadiazine, Sulfamerazine, Sulfamethazine, Sulfacetamide and Sulfisoxazole that show in vitro inhibition effect for BChE were determined as 3,397 mM, 3,364 mM, 5,414 mM, 2,717 mM, 2,235 mM respectively. Standard deviations and mean Ki constants for the same samples were recorded as 0,815±0,098 mM, 3,104±0,736 mM, 2,763±0,055 mM, 0,923±0,191 mM, 0,582±0,143 mM respectively. While the inhibition types showed competitive inhibition in Sulfadiazine, Sulfacetamide and Sulfisoxazole, Sulfamerazine and Sulfamethazine showed noncompetitive inhibition effect.
