Effects of Okra (Abelmoschus esculentus) SEED extract on lipopolysaccharide-induced rat LUNG injury

Abstract (EN)
Background: Pediatric Acute Respiratory Distress Syndrome(PARDS) is one of the common causes of morbidity and mortality in pediatric intensive care patients. Although the etiopathogenesis of the disease has been largely clarified, there is still no effective treatment available. Although it is a clinical entity that may originate from different etiologies, the common feature of the disease is that excessive inflammatory response and increased oxidative load cause diffuse lung injury. Abelmoschus esculentus(AE) has antioxidant and anti-inflammatory properties. In our study, we aimed to investigate the effect of AE on acute lung injury. Material and Methods: A total of 40 male Winstar rats were randomly divided into 5 groups. The groups were arranged as control group (K), lipopolysaccharide group (L), AE group (AE), AE + L group (AE+L), AE + L + dexamethasone (AE+L+D) group. AE was injected at a dose of 600 mg/kg/day via gavage for 7 days, L 5 mg/kg was injected intraperitoneally on the 7th day as a single dose, and D 1,5 mg/kg was injected intraperitoneally once on the 7th day. Samples taken from rats were analyzed biochemically and histopathologically. TNF-α, IFN-γ, IL-10, angiopoietin-2, thiol and disulfide levels were analyzed in the samples taken from the serum. Lung samples were stained with hematoxylin-eosin and examined microscopically.. Results:Biochemically, TNF-α, IFN-γ, IL-10, angiopoietin-2, thiol and disulfide levels were investigated. Total thiol values were found to besignificantly higher in the L+AE and L+AE+D groups compared to the L group (p:0.004). IFN-γ values weresignificantly lower in the L+AEand L+AE+D groups compared to the L group (p<0.001 ve p<0.001 respectively). IL-10 values were found to be statistically significantly higher in the L+AE and L+AE+D groups compared to the L group (p=0.025 ve p<0.001, respectively). Compared to the L group, in the L+AE and L+AE+D groups, hemorrhage scores (p=0.006 and p=0.011, respectively), congestion scores (p=0.006 and p<0.001, respectively), intraalveolar macrophage scores (p=0.001 and p<0.001, respectively) were statistically significanly low.On the other hand, alveolar wall thickness and inflammation scores were significanly low only in the L +AE+D group compared to the L group (p<0.001 and p=0.001, respectively). Conclusion: AE has a protective effect against acute lung injury caused by oxidative stress and inflammation induced by lipopolysaccharide.
