Dr. Mehtap KUTLU2005, 64 sayfaIn this study, three different 9-substitüe phenantrene derivatives that were synthesized to beused as basic matter for drug were tested for mutagenic potency in strains TA 98 and TA 100 ofSalmonella typhimurium by using Salmonella Mutagenity Test. Therefore, both test strains weretested in the absence or presence of S9 metabolic activation, for five different doses of each testsubstances in two paralel independent experiments. Results were evaluated by calculating avarageof two experiments. At the absance of S9, 4-nitro-o-fenilendiamine (NPD) was used as a positivecontrol for TA 98, sodyum azid (AZD) was used as a positive control for TA 100. At the presence ofS9, (2-AP) 2-Amino-Fluorene was used as a positive control for both test strains. Solvent DimetylSulphoxid (DMSO) and spontane control groups were used as a negative control groups data. Sothat, 9-[4,5-di-(4-methoxyphenyl)-2H-imidazol-2-il]- phenanthrene was found to be mutagenic forTA 98 in the absence and presence of metabolic activation. It was also mutagenic in TA 100 withmetabolic activation. 9[2H-benzothiazole-2-il]-phenanthrene caused frameshift mutagenicity in theabsence of metabolic activation. 9-[1H-phenanthro-(9,10-d)-imidazol-2-il]-phenanthrene causedframeshift mutation with and without S9. This compound was non-mutagenic for TA 100.Somutagenic effecet may be depend on the metabolic activation.Key words: Ames Test, Phenanthrene Derivatives, Mutagenecity, MetabolicActivation, Polisiclic Aromatic Hydrocarbons
