Abstract (EN)
This research was carried out at the Field Crops Central Research Institute, Biotechnology Research Center, Yenimahalle campus in 2021 with the aim of increasing the regeneration rate in obtaining doubled haploid plants by anther culture method in some barley populations. In our study where three different hybrid populations (Tokak 157-37 (X) Aida (X) Sayım 40, Sayım 40 (X) Tokak 157-37, Javelin (X) Alena) were used as materials, plant explant (lower-middle-upper parts of barley spike), two different temperatures (+4-+28 ˚C) and Cubic Sulfate (CuSO4) concentrations (control-1-5-10 mg/L) were used as variables in pretreatment; anther culture, haploid plant production and regenerated plants as variables in pretreatment. With the variable pretreatments, 1 molar of solid mannitol medium was used first, followed by callus formation, callus development, and rooting media, respectively. After haploid anthers (with n chromosome number) were transferred to the first medium under sterile conditions, callus (the interval between anther and callus was 3-4 weeks), green callus, shoot, and rooted plantlet formations were observed, respectively. Afterward, they were kept at +4˚C for 30 days to meet the need for vernalization. Hybrid populations gave different results in terms of callus formation, number of shoots, and regenerated plants. This situation also revealed the importance of genotypes in the study.
