In vivo investigations of some plant extract effects on mast cells

Abstract (EN)
In this study, 20 female and male Wistar rats (Rattus rattus norvegicus) were used. Animals were divided into three groups: seven of them is experiment, seven of them is positive control and six of them is negative control animal. Experiment groups of rats have been injected the oregano (Origanum onites L.) volatile oils, positive groups of rats have been injected the olive oil and negative groups of rats have been injected the serum physiologıcal intraperitoneally. In this study we particularly investigated the effect of oregano on locations, morphology, degranulation of mast cells in skin containing connective tissue-type mast cells (CTMC) and mucosal-type mast cells (MMC) on stomach fungal section, and at the same time on staining of mast cell mediators. From mast cell mediators have been studied to determine heparine, histamin and seratonin on mast cells staining via specific stains using conventional histological methods, and obtaining findings have been compared with each other.In respect of obtaining findings, mast cells in groups injected oregano volatile oil and only olive oil have been determined to increase as numerically and to be localize in connective tissue in comparison with negative control groups. Also, mast cells have shown morphological and chemical differences; experiment groups have degranulised and granulised, positive control groups have only granulised . Furthermore, we determine that mast cells in stomach fungus perform their function secreting histamine and mast cells in skin secreting heparin. Finally, we think that oregano could use in medicine industry as raw material using advanced techniques in further studies taking into consideration that mediators secrete from mast cells effect positively on digestive and immune systems.Keywords: Origanum onites L., Mast Cell, Skin, Stomach, Mast Cell Mediators
