Abstract (EN)
The aim of this study is to examine whether some drug active ingredients have inhibitory and activator effects on lipase enzyme activity. Lipases (Triacylglycerol acylhydrolase; EC 3.1.1.3) are capable of converting to diand monoacylglycerols and glycerol by catalyzing the hydrolysis of triacylglycerols, hydrolyzing triacylglycerols at the lipid-water interface, and synthesizing ester bonds in anhydrous medium. It is known that the lipolytic activity of the lipase enzyme takes place at the enzyme lipid interface. It is known that obesity, which has been a serious health problem throughout the world and in our country in recent years, is a pioneer in the formation of many diseases such as hypertension, diabetes (Type II-diabetes mellitus), gallstones, respiratory dysfunction and cancer. Obesity is tried to be treated with classical methods depending on diet and exercise. However, the lost weight is regained after a while. Therefore, pharmacological treatments are recommended for long-term preservation of weight loss with diet, lifestyle changes. For this reason, orlistat used inhibits gastrointestinal lipases and reduces the absorption of fatty acids in the small intestine and ensures their excretion from the body through the large intestine. Along with this mechanism, pancreatic lipase inhibitors prevent the storage of fat in the body and cause weight loss. The most important lipase inhibitors are orlistat and its hydrogenated derivative, lipstatin. In some studies, it has been observed that naturally sourced ingredients such as flavonoid, hesperedin, stilben, catechin and terpenoid derivatives can also be used as lipase inhibitors. In addition, bioautographic methods combined with chromatographic techniques to quickly screen naturally sourced lipase inhibitors can qualitatively show pancreatic lipase activity for each component of naturally sourced extracts composed of multiple components.
