Abstract (EN)

Since legumes are an essential source of protein, they have a privileged place, especially in developing countries. Bean grains from the legumes family are consumed extensively in the world and in our country, due to their rich carbohydrate and protein content, as well as their high calories, as well as being deliciousness. Bacterial bean diseases cause both low yield and uneconomical use of chemical pesticides. Breeding studies using marker-assisted selection (MAS) is an alternative method for developing resistant bean varieties in the fight against diseases. In this study, it was aimed that determine of resistance gene profiles against halo blight (Pseudomonas syringae pv. phaseolicola) and common leaf blight (Xanthomonas axonopodis pv. phaseoli) diseases in beans (Phaseolus vulgaris L.) in GKTAEM (Eskişehir Transitional Zone Agricultural Research Institute) crossbreeding studies, using specific molecular markers. Seventy-two advanced lines including pole and dwarf genotypes of bean varieties developed by crossbreeding were used. DNA isolations from bean samples were performed using a commercial kit. PZR (polymerase chain reaction) amplifications were performed using four SCAR (sequence characterized amplified region) markers against halo blight (HB: HY) and seven SCAR markers against common leaf blight (CLB: AYY). PZR products were run and visualized in agarose gel electrophoresis with ethidium bromide. Results of the resistance gene profile analysis showed that the 20-SBD-25, 20-SBarBD-12, and 20-SD-10 bean lines containing more genes than the others stood out. As a result of the study, the data of the resistance gene profiles of seventy-two bean lines can be used directly within the scope of MAS. This study will contribute to the parental selection of breeding studies for the development of bean bacterial diseases resistant varieties in our country.
