Analysis of factors affecting metabolism of trehalose and glycogen metabolisms in some yeast species

Abstract (EN)

Trehalose and glycogen are common carbohydrates in many organisms including bacteria, yeasts, plants and invertebrates. These glucose stores also have a regulatory role in the control of glycolytic flux. Infact, trehalose and glycogen accumulate in response to various environmental stress conditions to protect cellular components.In this study, the trehalose and glycogen contents of C. albicans, C. rugosa, M. fructicola, P. stipitis, Y. lipolytica, Z. bailii, Z. rouxii and S. cerevisiae were investigated in response to different stress condition such as nutritional stress, oxidative stress, heat shock and metal stress. There were similarities and differences between these yeast species in response to stress conditions regarding their trehalose and glycogen contents. It is found that the heat shock results in a significant level of glycogen and trehalose accumulation in these yeasts. Moreover, high levels of glycogen accumulations were detected in C. rugosa in response to all stress conditions. It is determined expression of TPS1 and GSY1 in C. rugosa, C. albicans, S. cerevisiae with RT-PZR. Accumulation of trehalose and glycogen, which was determined by enzymatic methods was compared with that determined by molecular techniques. C. albicans GSY1 is induced (78,13) by cadmium stress according to stress-free condition. In this research, possible metabolic connections between pseudohypeal development and reserve carbohydrate metabolism are investigated by biochemical methods in yeasts used in this research. Pseudohypeal development was followed microscopically in different time intervals. In addition, we have also identified the species at yeast samples obtained from vagina as C.glabrata with sequencing analysis.Keywords: Yeast, stress, trehalose, glycogen, RT-PCR, sequencing analysis
