Synthesis and biological activities of some new 4,5-Dihydro-3(2H) -pyridazinone derivatives

Alzheimer is a neurodegenerative disease which decreases the quality of life of people, common cause of mortality of geriatric population and its pathogenesis is not known exactly. As the worldwide, it's incidence tends to increase in Turkey also. Although there are hypthesis trying to explain pathophysiology of the disease, none of them cannot determine the exact reason. In this way, the mechanisms of medicines that are using in clinical practice are based on cholinergic hypothesis. The disadvantages of treatment is the interaction of existing medicines of the patients with the chronic disease as well as the many side effects of medicines using in treatment. Pyridazinone that have proven itself anticholinergic activity is involved in many studies. There is no Pyridazinone ring system in the medicines that are currently using in clinical treatment. In this study, we are synthesized new some 4,5-Dihydro-3 (2H) -pyridazinone derivatives for developing a new anticholinergic agent. The new molecules synthesized by reacting the 3(2H)-pyridazinone ring with acid anhydrides. The structures of the compounds were determined by elementel analyses and spectral data. The anticholinergic activities of the compounds were tested on butyrylcholinesterase and acetylcholinesterase enzymes. We found moderate levels of A1, A3 compounds and low level of anticholinergic effects in A2, A4 and A5 compounds.
